Drug effects mediated by the immune system are evaluated by quantification of secreted soluble factors in mouse serum/plasma or by measuring phenotypical or function-related parameters of mouse immune cells.
Changes in blood / serum parameters:
Characterisation of mouse immune cells:
T-, B-, NK- and monocytic/dendritic cells are isolated from blood or spleen
using Magnetic Cell Separation (Miltenyi). Purity control of isolated immune
cells is performed by FACS analysis. Functional parameters (basal and
stimulation capacity, intracellular level of IL-2, TNFα IFNγ or other cytokines)
are measured by FACS analysis.
